SEGREGATION ANALYSIS OF BIOACTIVE GENOTIPE AND MORPHOLOGY IN RICE F2 THE CROSS RESULT BETWEEN KETAN HITAM AND CIHERANG

Rice plants can be grouped into 2 types, namely pigmented and non-pigmented rice. One example of pigmented rice is brown rice and black rice. Pigmented rice is known as a source of antioxidant compounds including flavonoids, anthocyanins and phenolic compounds. The aim of this study was to analyze the segregation of the results of crosses between Ketan Hitam and Ciherang rice varieties through analysis of morphological characters and bioactive genotypes using SSR molecular markers. The research methods used were plant preparation and maintenance (preparation of planting material, tillage, planting, fertilization, irrigation, weeding, and harvesting) and bioactive genotype analysis (DNA extraction, PCR work process, electrophoresis and UV transluminator observation). PCR analysis used SSR markers with RM 346, RM 316, RM 228, and RM 339 primers. Analysis of the data used was quantitative frequency distribution analysis with IRRI guidance. The segregation of morphological characters in the F2 population from crosses between Black Sticky Rice and Ciherang is still high, which can be seen from the character segregation pattern on plant height, number of tillers, productive tillers and weight of 1000 seeds which are controlled by many genes and influenced by environmental factors in them because they are not meet the Mendelian genetic ratio or deviation. The morphological forms of the crosses tend to be similar to the female elders or Black Sticky Rice. Meanwhile, the PCR results using SSR markers showed that RM 346, RM 339 and RM 228 could validate the entire sample used, both the parent of the cross and the F2 of the cross, while RM 316 could not validate all the DNA bands in the research sample.
